Scientific Background

RelB is a pleiotropic transcription factor belonging to the Rel-like domain protein family, forming NF-kappa-B heterodimers (e.g., RelB-p50 and RelB-p52) that function as transcriptional activators by binding to kappa-B sites in target gene DNA, regulating processes like inflammation, immunity, and cell differentiation. RelB does not directly associate with DNA, RELA/p65, or REL but stimulates promoter activity in the presence of NFKB2/p49. As part of the NUPR1/RELB/IER3 survival pathway, RelB may confer remarkable resistance to cell stress (e.g., starvation or gemcitabine treatment) in pancreatic ductal adenocarcinoma. Additionally, RelB represses the transcriptional activity of the CLOCK-BMAL1 heterodimer independently of CRY1/CRY2, with enhanced repression observed with NFKB2/p52. RelB is essential for the maturation and function of T and B lymphocytes.
