SP1 is a transcription factor capable of activating or repressing transcription in response to physiological and pathological stimuli. It binds with high affinity to GC-rich motifs and regulates the expression of numerous genes involved in processes such as cell growth, apoptosis, differentiation, and immune responses. SP1 is highly regulated by post-translational modifications (e.g., phosphorylation, sumoylation, proteolytic cleavage, glycosylation, and acetylation). It also binds the PDGFR-alpha G-box promoter and may modulate cellular responses to DNA damage while participating in chromatin remodeling. SP1 plays an essential role in regulating FE65 gene expression and, in complex with ATF7IP, maintains telomerase activity in cancer cells by inducing TERT and TERC gene expression. Isoform 3 exhibits stronger transcriptional activation than isoform 1 and positively regulates the core clock component BMAL1. Additionally, SP1 facilitates the recruitment of SMARCA4/BRG1 to the c-FOS promoter. By similarity, SP1 protects cells against oxidative stress following brain injury by regulating RNF112 expression.
