Scientific Background

Actinin α2 is the principal structural protein of cardiac Z-discs, functioning as an F-actin crosslinking protein that connects actin filaments to TTN filaments and maintains Z-disc integrity (by similar). Unlike Actinin α3 which is exclusively expressed in fast-twitch fibers, Actinin α2 is ubiquitously present in all skeletal and cardiac muscles, participating in mechanotransduction and metabolic regulation through interactions with various Z-disc proteins (e.g., titin, myotilin). Mutations in Actinin α2 are associated with hypertrophic and dilated cardiomyopathies, while the R577X polymorphism of Actinin α3 influences muscle power and endurance performance (by similar).
