TATA Box Binding Protein (TBP) is a core component of the TFIID basal transcription factor complex, playing a pivotal role in the initiation of RNA polymerase II (Pol II)-dependent transcription. TBP recognizes and binds promoters with or without a TATA box, facilitating the assembly of the pre-initiation complex (PIC). The TFIID complex comprises TBP and TBP-associated factors (TAFs), including TAF1 to TAF13, and is structurally divided into three modules: TFIID-A, TFIID-B, and TFIID-C, with TBP forming the TFIID-A module alongside TAF3 and TAF5. During PIC assembly, TBP binds and bends promoter DNA. Additionally, TBP is part of a BRF2-containing transcription factor complex that regulates RNA polymerase III-mediated transcription, and it is a component of the SL1/TIF-IB transcription factor complex involved in RNA polymerase I-dependent PIC assembly. SL1 also stabilizes the binding of nucleolar transcription factor 1/UBTF to rDNA.
