TGF β (Transforming growth factor beta-1 proprotein) is the precursor of Latency-associated peptide (LAP) and Transforming growth factor beta-1 (TGF-beta-1) chains, forming the regulatory and active subunits of TGF-beta-1, respectively. It maintains TGF-beta-1 in a latent state via non-covalent interaction with milieu molecules (e.g., LTBP1, LRRC32/GARP, LRRC33/NRROS) (By similarity). Integrins (ITGAV:ITGB6 or ITGAV:ITGB8) distort LAP to release active TGF-beta-1, which then signals through TGFBR1/TGFBR2 receptors. This multifunctional protein regulates cell growth/differentiation, immunity, bone remodeling (By similarity), and exhibits concentration-dependent roles in T-cell differentiation (promoting Treg at high levels or Th17 with IL-6/IL-21 at low levels) (By similarity).
