THAP1 is a DNA-binding transcription regulator that controls endothelial cell proliferation and G1/S cell-cycle progression. It specifically binds the 5'-[AT]NTNN[GT]GGCA[AGT]-3' core DNA sequence and modulates the expression of pRB-E2F cell-cycle target genes, including RRM1. As part of the THAP1/THAP3-HCFC1-OGT complex, it is essential for regulating RRM1 transcriptional activity. Additionally, THAP1 may exhibit pro-apoptotic activity by enhancing both serum-withdrawal and TNF-induced apoptosis.
