Scientific Background
URI1 is a protein involved in gene transcription regulation, acting as a transcriptional repressor in concert with the corepressor UXT to modulate androgen receptor (AR) transcription. It may function as a tumor suppressor in prostate cancer cells by repressing AR-mediated gene transcription and inhibiting anchorage-independent growth. In ovarian cancer cells, URI1 is essential for cell survival and, together with UXT, associates with chromatin at the NKX3-1 promoter region. Additionally, URI1 antagonizes transcriptional modulation mediated by the hepatitis B virus X protein. Under normal growth conditions, URI1 plays a central role in maintaining S6K1 signaling and BAD phosphorylation, protecting cells from the detrimental effects of sustained S6K1 signaling. The URI1-PPP1CC complex serves as a core component of a negative feedback mechanism that counteracts excessive S6K1 survival signaling to BAD in response to growth factors. URI1 also mediates the inhibition of PPP1CC phosphatase activity in mitochondria and coordinates nutrient-sensitive gene expression regulation in an mTOR-dependent manner. By similar function (by similar): URI1 appears to act as a scaffolding protein capable of assembling a prefoldin-like complex containing PFDs and proteins involved in transcription and ubiquitination.
