Flk-1/VEGFR2 is a tyrosine-protein kinase that functions as a cell-surface receptor for VEGFA, VEGFC, and VEGFD. It plays a pivotal role in regulating angiogenesis, vascular development, vascular permeability, and embryonic hematopoiesis by promoting endothelial cell proliferation, survival, migration, and differentiation, as well as reorganizing the actin cytoskeleton. Isoforms lacking a transmembrane domain (e.g., isoform 2 and isoform 3) may act as decoy receptors for VEGFA, VEGFC, and/or VEGFD. Specifically, isoform 2 negatively regulates VEGFA- and VEGFC-mediated lymphangiogenesis by sequestering free ligands and blocking their binding to FLT4. The receptor modulates FLT1 and FLT4 signaling through heterodimer formation. Ligand binding to isoform 1 triggers multiple signaling cascades, including PLCG1 activation (generating diacylglycerol and inositol 1,4,5-trisphosphate), protein kinase C activation, MAPK1/ERK2, MAPK3/ERK1, MAP kinase pathway, and AKT1 pathway. It also mediates phosphorylation of PIK3R1 (regulatory subunit of phosphatidylinositol 3-kinase), PTK2/FAK1 activation, and VEGFA-induced NOS2/NOS3 expression to stimulate nitric oxide (NO) production in endothelial cells. Known phosphorylation substrates include PLCG1, FYN, NCK1, NOS3, PIK3R1, PTK2/FAK1, and SRC.
