YTHDF1 is a protein that specifically recognizes and binds to N6-methyladenosine (m6A)-containing mRNAs, regulating their stability and thus influencing gene expression. m6A is a modification found at internal sites of mRNAs and some non-coding RNAs, playing a role in mRNA stability and processing. YTHDF1 promotes the degradation of m6A-containing mRNAs by interacting with the CCR4-NOT complex (By similarity). The YTHDF paralogs (YTHDF1, YTHDF2, and YTHDF3) share m6A-containing mRNA targets and function redundantly in mediating mRNA degradation and cellular differentiation. In the hippocampus, YTHDF1 facilitates learning and memory formation by binding to m6A-modified neuronal mRNAs. It also regulates axon guidance through interaction with m6A-containing ROBO3 transcripts. In myeloid dendritic cells, YTHDF1 acts as a negative regulator of antigen cross-presentation, limiting anti-tumor responses by reducing tumor-antigen cross-presentation efficiency during tumorigenesis. Additionally, YTHDF1 undergoes liquid-liquid phase separation upon binding to mRNAs with multiple m6A-modified residues, promoting the formation of membraneless compartments such as P-bodies or stress granules (By similarity). These mRNA-YTHDF complexes then partition into various endogenous membraneless compartments.
