Scientific Background
ZAP70 is a tyrosine kinase essential for regulating adaptive immune responses. It controls motility, adhesion, and cytokine expression in mature T-cells, thymocyte development, and contributes to primary B-lymphocyte activation. Upon TCR stimulation by APCs, phosphorylation cascades recruit ZAP70 to doubly phosphorylated CD247/CD3Z via ITAM motifs, localizing it to activated TCR and releasing its autoinhibited conformation—further stabilized by LCK-mediated phosphorylation. ZAP70 then phosphorylates adapter proteins LAT and LCP2, initiating signaling cascades that drive lymphokine production, T-cell proliferation, and differentiation. It also modulates T-cell cytoskeleton dynamics for effector delivery to APCs and regulates TCR internalization/degradation via interactions with CBL, SLA, and SLA2, thereby controlling TCR surface expression for activation modulation. During thymocyte development, ZAP70 promotes survival and cell-cycle progression in CD4/CD8 double-negative stages. By similar mechanisms, it may support primary B-cell formation/activation through BCR signaling.
