Scientific Background

cIAP1 is a multi-functional protein that regulates caspases and apoptosis, while also modulating inflammatory signaling, immunity, mitogenic kinase signaling, cell proliferation, invasion, and metastasis. It functions as an E3 ubiquitin-protein ligase to regulate NF-kappa-B signaling, acting as a positive regulator of the canonical pathway while suppressing constitutive activation of the non-canonical pathway. Its E3 ligase targets include RIPK1, RIPK2, RIPK3, RIPK4, CASP3, CASP7, CASP8, TRAF2, DIABLO/SMAC, MAP3K14/NIK, MAP3K5/ASK1, IKBKG/NEMO, IKBKE, and MXD1/MAD1. Additionally, it can act as an E3 ligase in the NEDD8 conjugation pathway, inactivating effector caspases via neddylation. cIAP1 is a key regulator of innate immune signaling through Toll-like receptors (TLRs), Nod-like receptors (NLRs), and RIG-I-like receptors (RLRs), collectively known as pattern recognition receptors (PRRs). It prevents spontaneous ripoptosome formation—a multi-protein complex capable of killing cancer cells in caspase-dependent or -independent manners—by ubiquitinating RIPK1 and CASP8. Furthermore, cIAP1 enhances the transcriptional activity of E2F1 and plays a role in cell cycle modulation.
