eIF4A3 is an ATP-dependent RNA helicase involved in pre-mRNA splicing as a core component of the spliceosome. It serves as a core member of the splicing-dependent multiprotein exon junction complex (EJC), which is deposited at mRNA splice junctions. The EJC is a dynamic structure consisting of core proteins and transiently associated peripheral factors, marking exon-exon junctions and influencing downstream mRNA metabolism processes such as nuclear export, subcellular localization, translation efficiency, and nonsense-mediated mRNA decay (NMD). Its RNA-dependent ATPase and helicase activities are induced by CASC3 but inhibited by the MAGOH-RBM8A heterodimer, stabilizing the ATP-bound EJC core on spliced mRNA. eIF4A3 enhances translation of spliced mRNAs post-80S ribosome formation and binds mRNA sequence-independently 20-24 nucleotides upstream of exon-exon junctions, exhibiting higher RNA affinity in the ATP-bound EJC state. It also modulates splicing of apoptotic genes like BCL2L1/Bcl-X, specifically suppressing proapoptotic isoforms (e.g., Bcl-X(S)), a function distinct from canonical EJC assembly. Additionally, it plays a role in craniofacial development.
