Scientific Background

p40/p63 functions as a sequence-specific DNA-binding transcriptional activator or repressor. Its isoforms contain varying transactivation and auto-regulating transactivation-inhibiting domains, resulting in isoform-specific activities. Isoform 2 activates RIPK4 transcription. In response to genotoxic insults and activated oncogenes, p40/p63 may cooperate with TP73/p73 to initiate p53/TP53-dependent apoptosis. It participates in Notch signaling, likely by inducing JAG1 and JAG2 expression. p40/p63 plays a crucial role in regulating epithelial morphogenesis, where the ratio of DeltaN-type to TA*-type isoforms may govern epithelial stem cell compartment maintenance and initiation of epithelial stratification from undifferentiated embryonal ectoderm. This protein is also essential for limb formation from the apical ectodermal ridge and activates transcription of the p21 promoter.
