Beta III tubulin is abundant in the central and peripheral nervous systems (CNS and PNS) where it is prominently expressed during fetal and postnatal development. As exemplified in cerebellar and sympathoadrenal neurogenesis, the distribution of beta III is neuron-associated, exhibiting distinct temporospatial gradients according to the regional neuroepithelia of origin. However, transient expression of this protein is also present in the subventricular zones of the CNS comprising putative neuronal and/or glial precursor cells, as well as in Kulchitsky neuroendocrine cells of the fetal respiratory epithelium. This temporally restricted, potentially non-neuronal expression may have implications in the identification of presumptive neurons derived from embryonic stem cells.

TUBB3, the class III β tubulin or Tuj1, is selectively expressed in testis and neurons of the central and peripheral nervous system. It has been widely used as a marker for neurons. Aberrant expression of TUBB3 has also been found in various tumors of non-neural origin and can be used as a biomarker for cancer aggressiveness and a marker for the tendency to respond poorly to chemotherapy. This antibody is specific to TUBB3 but not cross-react with other tubulin isoforms.

Tubulin, beta 3, also known as TUBB3. Tubulin is the major constituent of microtubules. It binds two moles of GTP, one at an exchangeable site on the beta chain and one at a non exchangeable site on the alpha chain. Tubulin is a highly conserved protein with a molecular weight of ~50 kD. Microtubules play key roles in chromosome segregation in mitosis, intracellular transport, ciliary and flagellar bending, and structural support of the cytoskeleton. The two main tubulin isoforms, alpha and beta tubulin, are usually products of separate genes. The beta tubulin family includes six expressed genes that produce the polypeptide isoforms known as Classes I through VI, each of which have a distinct pattern of expression. Class III beta tubulin is found in neurons and mammalian testis cells and is widely used as a neuronal marker in developmental neurobiology, neoplasia, and stem cell research. Class III beta tubulin expression in neuronal and neuroblastic tumors is differentiation dependent, and its expression in certain non-neuronal neoplasms has been associated with poor prognosis and/or resistance to chemotherapy.
