The monoclonal antibody ER-MP20 specifically reacts with mouse macrophage precursor cells in the mid-stage of their development (late CFU-M, monoblasts and monocytes). The antigen is a 14 kD surface protein which is very similar to Ly-6C and may be analogous to human CD59. It is inducible by IFN-alpha, IFN-beta and IFN-gamma. In tissue sections, the antigen is found on macrophage precursor subpopulations. In the bone marrow and hemopoietic islands of the lymphoid organs, and in the spleen. Activated macrophages in inflammatory tissues also express the ER-MP20-related antigen. The monoclonal antibody ER-MP20 has been raised after immunization of rats with mouse macrophage cell lines and reacts with mouse macrophage precursor cells. The monoclonal antibody also identifies activated macrophages in inflammatory tissues where the simultaneous use of the murine pan-macrophage marker BM8 (anti-F4/80) is recommended. In combination with an anti-mouse CD31/PECAM-1 antibody, ER-MP20 can be used to evaluate the cellular composition in murine bone marrow (e.g. using flow cytometric analysis). ER-MP20 also detects a wide range of endothelial cells.

The monoclonal antibody ER-HR3 is reactive to the majority of mouse monocytes and a subset of mature resident macrophages, especially those located in hemopoietic organs. Whereas all other leukocytes are ER-HR3 negative, up to 70% of circulating monocytes stain positive with this monoclonal antibody. Furthermore, about 30% of bone marrow cells express the antigen. Other ER-HR3 positive macrophages can be found in the splenic red pulp, in the mesenteric lymphoid paracortex, interfollicular areas of Peyer’s patches and bone marrow. Few ER-HR3 positive cells are observed in the thymic cortex and the connective tissues of the gastro-intestinal tract, the dermis and the renal medulla. Epidermal Langerhans cells also express the antigen. The monoclonal antibody ER-HR3 can be used to identify and localize a very distinct mature tissue macrophage subpopulation found in various mouse organs and its distribution pattern is unique compared to that from other macrophage-specific antibodies. The antibody is especially suitable for ontogenic studies because ER-HR3 positive macrophages are closely related to hemopoietic islands, especially at erythropoietic sites.

The monoclonal antibody ER-MP12 recognizes mouse platelet endothelial cell adhesion molecule-1 (PECAM-1;CD31). PECAM-1 is a 130kDa glycoprotein involved in- cell adhesion and a signalling receptor that is expressed on hematopoietic and endothelial cells. It is a member of the immunoglobulin (Ig)-superfamily of cell adhesion molecules. The type I transmembrane protein is composed of an extracellular region of six Ig-like homology domains, a 19-residue transmembrane domain, and a 118 residue cytoplasmic tail and is differentially glycosylated involving N-linked and O-linked glycosylation sites. The signalling of PECAM-1 is based on 2 immuno receptor tyrosine inhibitory motifs (ITIMs) in its cytoplasmic domain. These ITIM domains can serve as docking sites for signalling molecules such as protein tyrosine phosphatases, and ligation of PECAM-1 can induce phosphorylation of tyrosine- and serine/threonine-residues of these intracellular regions eventually leading to activation of signalling pathways. The biological characteristics- of PECAM-1 in these pathways and cellular adhesion have been mapped to specific regions of the PECAM-1 molecule. PECAM-1 is abundantly expressed on endothelial cells, where it is a major constituent of the endothelial cell intercellular junction in confluent vascular beds, making it a commonly used endothelial marker. Furthermore, it is expressed on most cells of the hematopoietic lineage including platelets, monocytes, neutrophils, and lymphocyte subsets. Besides adhesive properties PECAM-1 is an efficient signalling molecule. The protein is involved in vascular biology including- angiogenesis, platelet function, and thrombosis, mechanosensing of endothelial cells, and regulation of multiple stages of leukocyte migration through venular walls. PECAM-1 is also a regulator of inflammatory responses. It has been shown to serve a variety of pro-inflammatory and anti-inflammatory functions. Pro-inflammatory functions of PECAM-1 include the facilitation of leukocyte transendothelial migration and the transduction of mechanical signals in endothelial cells originating from fluid shear stress. Anti-inflammatory functions include the dampening of leukocyte activation, suppression of pro-inflammatory cytokine production, and the maintenance of vascular barrier integrity. Monoclonal antibody ER-MP12 reacts also with the major population of colony forming unit macrophage (CFU-M) precursor cells, with a subpopulation of pre CFU-M and monoblasts, and with precursor cells of granulocytes. The antibody can also be used, in combination with the monoclonal antibody ER-MP20, for identification of different stages of myeloid progenitor cell development.
